Confocal Microscopy Lab Confocal microscopy uses a laser that produces excitation light. This laser light reflects off of a dichroic mirror and then the laser light hits two mirrors that are mounted on motors. The mirrors then scan the laser light across the sample. Dye that is found in the sample then fluoresces (Weeks, 2003). Filamin was labeled with a red fluorescent label rhodamine (TRITC) and actin was labeled with the green fluorescent label fluoroscein (FITC) which was conjugated to the actin-binding fungal toxin phalloidin. The emitted light from the dyes passes back through the mirrors and passes through the dichroic mirror and is focuses into a pinhole. With confocal microscopy, a complete image of the sample is never seen. Only one point of the sample can be observed at a time. The amount of light that passes back through is detected by the microscope (Ladic, 1995). The intensity of the red light seen is proportional to the amount of filamin present and the intensity of the green ligh t seen is proportional to the amount of actin present in the sample of Drosophila melanogaster ovaries. Confocal microscopy is able to produce images that are very free from interference. The confocal pinhole allows the microscope to reject out of focus fluorescent light (Weeks, 2003). This means that the image comes from a thin section of the ovary sample. Many thin sections will be scanned through the sample; this allows a clean three dimensional image to be made. A confocal microscope has a few advantages over regular optical microscopes. Use of Medium & Elements :: Drama

Use of Medium & Elements During performing scenes from A Dolls House by Henrik Ibsen. We used two different types of Styles. Naturalistic and Non Naturalistic. We worked in groups on each style. Naturalistic is a form of drama which is realistic. Not many strategies can be used doing this form. Non Naturalistic drama can contain many explorative strategies, such as, Hot seating, Still image, and thought tracking. The first style we worked on was Non Naturalistic. We chose to the scene where Krogstad confronted Nora. We made Krogstad two people to try and show Krogstad's power over Nora. We alternated the lines between two actors. (Use of spoken language) Krogstad was wrapped in a big black blanket to represent a black hole that Nora had fallen into. A hole of lies and deceit, towards Torvald. Nora was dressed in white to represent her innocence that she thought she had. We used these colours for symbolism in this production. This is a drama medium. Using symbolism showed me how different art forms react with other art forms. (E.g. colour and script) The script changed for me from then on. The white and black was a contrast that is widely used in the world. White is used for angels black is used for hell. White is used for a wedding black is used for a funeral. White is day. Black is night. People are scared of the night but never the day. During this scene we used space wisely. Nora moved backwards as Krogstad moved forwards. Using movement Krogstad would fly the black blanket around over Nora head trying to suck her under it. Nora would back away though trying to resist the dark side. (The outside of the house). Towards the end of scene where Krogstad dominates Nora and gets the truth out of her They exchanged blanket. Nora is now wrapped in black, which symbolises the outside world and the lies and deceit. It shows the viewer that Nora has no way of escaping what she has done. Using Non- Naturalistic drama to produce a scene from A Dolls House helped me understand the script a bit more. It certainly helped me figure out how scary the outside world was for Nora and Torvald. The symbolism in the blankets were a great help also in portraying how Nora had lied and it had caught up with her. Carrying the black blanket did help me imagine what the outside world must be like for the Helmer household. Holding the blanket and moving it closer to Nora who was a big sheet of white pure innocence felt quite evil yet

Play Observation Essay

Kiahi a boy of about 3 years old was playing alone stood on a small hill of dirt , at first it looked like her was just looking at the other kids play. When I kept looking at him to see if he was just going to continue to doing nothing, but watch the other children I noticed that he was standing there like a solider tall and with determination on his face. With his hand to his forehead looking about as the other children ran around. After a while I see that he is pointing to his class mates at random and looks to be giving directions, more like orders, of some sort, although no one is paying attention. After giving off orders in a solider like manner he nods his head, as though satisfied with himself and his team though it was just him playing, and pulls out some kind of black/sliver rectangular key chain object out of his shirt pocket. He clicks on it, as if it had buttons, brings it up to his ear and talks and nods, he’s using it as a cell phone. After he is done he clicks i t again to end the call and puts it back to his pocket. Then he just runs off the hill and goes plays at the play structure, as if what he just did never happened. At first I thought Kiahi was engaging in unoccupied play, because to me it looked as if he was doing nothing at all but just looking at what everyone else was doing. As I kept observing him it looked to him that his was doing solitary play. Playing out a role of someone with authorization giving out orders, and taking calls. It was solitary because no one else was engaging with him in is play even though he pointed out to other kids when giving off orders no one paid him attention. Sophia a girl of about 4 years old with a temperament of a being shy and quite, wasn’t playing with anybody else. She was reluctant to join anybody for play. After a while Sophia looks to have acquired a play mate Elena a soft spoken 3 years old, though in some occasions I did hear her be a little louder towards other kids, they are playing in the sand box. It looks like they are playing kitchen; cooking, serving, cutting and cleaning. Throughout the play not a word was spoken, but they seemed to communicate in their own silent way. After a while Sophia and Elena moved to play with the balls, when Sophia would turn her back on the ball and it rolled away from her instead of calling out to Sophia to get the ball Elena runs after the ball and gives it back to Sophia in her hands and points to the baskets behind her. In a silent gesture to throw the ball at the basket. I found these  girls interesting because I’ve observed them for a while during the morning time when dropped off up until free play outside. I know for a fact that both girls speak Elena even yells at times when kids don’t pay attention for following the rules. Sophia is new to the pre-school program at Head Start and cries when her Mom, as Ms.Perla informed me, leaves her but just for a few minutes. It takes Sophia a few hours to get into play with actual words being spoken. Elena is a sweet little that separated from her little group of friends to play with Sophia and to me it looked as if Elena understood Sophia not wanting to speak, seeing as Elena ever attempted to say a single word to her. At first Sophia was engaging in solitary play, playing on her own on the play structure. In the sand box Sophia and Elena cooperative playing, even though they didn’t say a word, the worked together to prepare, cook and serve a meal and clean up after they were done. They had a common goal in playing kitchen. After that they played with the balls which turned to associative play still playing together but without a common goal because Sophia wasn’t as engaged as Elena in trying to play basketball.

Fast Food Restaurant Description

Scene in a Fast Food Restaurant. I push through the crowds of young people hovering outside the automatic doors of Burger King, kicking the empty paper cups and bags out of my way. Stepping inside, the first thing that hits me is the sound. It crashes over me, engulfing me, drawing me in. I step closer, into the midst of it. To my left sit a young couple, anxiously feeding their toddler chicken nuggets dipped in tomato sauce. The two year old cries and whines, putting his hand up to his mouth as if to say ‘no, no more. ’ The group of young people to my right are laughing, shouting and flirting.One of the boys has stolen a girl’s milkshake and she leans across her friends, giggling happily, to try and snatch it back. I can hear the radio playing faintly. The newest, noisiest dance track struggles to be heard in the room full of people, resembling a school canteen. As I make my way upstairs I pass a smartly dressed businessman, holding a brown bag containing a burge r, and his other hand to hold his drink. He has his mobile phone trapped between his ear and his shoulder and he jabbers away to his colleague about ‘redundancies’.An elderly woman, accompanied by two young, brightly dressed grandchildren, frowns at the man as she makes her way past, children in tow. The smell of the greasy, fatty burgers is overpowering now, and I can hardly breathe for the stench if freshly cooked French fries. They coat the floor, like a three-inch carpet, soft underfoot. I wonder why these restaurants even bother installing bins – nobody seems inclined to use them. Spotting no empty tables, I make my way back downstairs to order my food. I overtake the queue if people waiting for ‘veggie-burgers’ and order large fries and a chocolate milkshake.The young girl who serves me can’t be much older than myself, yet she looks older, more tired, world-weary. Her shoulder length hair hangs limp and greasy under her baseball cap, and her red t-shirt is stained with fat and fizzy drinks. The woman next to me has dropped her tray, and someone with a mop rushes to clean up the split cola, before anyone has a chance to fall in it. I smell the air, take a French fry out of the packet, pop it in my mouth and sigh. It tastes like grease, unhealthy and fattening.
